General Technique of Plant Tissue Culture
यह आदर्श रूप से 7 चरणों में किया जाता है:-
(It is ideally done in 7 stages: -)
1. कर्तोतक का चयन (Selection of Explant):-
• Explant (कर्तोतक):-
पौधे का वह भाग जिसे माध्यम पर संवर्धित करने के लिए चुना जाता है, कर्तोतक कहलाता है।
(The part of the plant which is selected for culturing on the medium is called explant.)
2. कर्तोतक का निर्जमीकरण (Sterilization of Explant):-
• कर्तोतक की सतह पर उपस्थित जीवाणुओं या कवकों के बीजाणुओं को नष्ट करने के लिए निम्न निर्जमीकारकों का उपयोग कर सकते हैं:-
(To destroy the spores of bacteria or fungi present on the surface of explant, the following sterilizing agents can be used:)
i. NaOCl = 1-1.4%
ii. Ca (OCl)2 = 9-10%
iii. H2O2 = 10-12%
iv. AgNO3 = 1%
v. Hg2Cl2 = 0.01 - 1%
3. काँच के बर्तनों का निर्जमीकरण (Sterilization of Glasswares):-
• काँच के बर्तनों का ओवन में शुष्क गर्म हवा के द्वारा निर्जमीकरण किया जाता है।
(Glass wares are sterilised by dry hot air in the oven.)
