Cyclic and non-cyclic photophosphorylation; Hill reaction; Oxygen evolution in photosynthesis (चक्रीय और अचक्रीय फोटोफॉस्फोराइलेशन; हिल अभिक्रिया; प्रकाश-संश्लेषण में ऑक्सीजन का निकलना):-
Light Reaction (Photochemical phase) [प्रकाश अभिक्रिया (प्रकाश-रासायनिक चरण)]:- इसमें प्रकाश अवशोषण, जल विपाटन, ऑक्सीजन का निकलना और उच्च ऊर्जा रासायनिक मध्यवर्तीयों (ATP और NADPH) का निर्माण शामिल है।
[It includes light absorption, water splitting, oxygen release, and formation of high-energy chemical intermediates (ATP & NADPH).]
a. The Electron Transport:-
·         जब PS II, लाल प्रकाश की 680 nm तरंग दैर्ध्य को अवशोषित करता है, तो इलेक्ट्रॉन उत्तेजित होते हैं और एक इलेक्ट्रॉन ग्राही पर स्थानांतरित हो जाते हैं।
(When PS II, absorbs 680 nm wavelength of red light, electrons are excited and transferred to an electron acceptor.)
·          इलेक्ट्रॉन ग्राही उन्हें इलेक्ट्रोन ट्रांसपोर्ट सिस्टम की एक श्रृंखला में पास करता है जिसमें साइटोक्रोम होते हैं।
(The electron acceptor passes them to a chain of electrons transport system consisting of cytochromes.)
·         रेडॉक्स विभव पैमाने के संदर्भ में इलेक्ट्रॉनों की यह गति डाउनहिल होती है।
(This movement of electrons is downhill, in terms of redox potential scale.)
·         इलेक्ट्रॉनों को PS I के वर्णकों को स्थानांतरित किया जाता है।
(The electrons are transferred to the pigments of PS I.)
·         इसके साथ ही, PS I में इलेक्ट्रॉन्स भी उतेजित हो जाते हैं जब वे लाल प्रकाश (700 nm) प्राप्त करते हैं और एक अन्य ग्राही अणु पर स्थानांतरित कर दिया जाता है जिसका रेडॉक्स विभव अधिक होता है।
[Simultaneously, electrons in PS I are also excited when they receive red light (700 nm) and are transferred to another accepter molecule having a greater redox potential.]
·         ये इलेक्ट्रॉन्स NADP+ के एक अणु पर डाउनहिल रूप से स्थानांतरित होते हैं। परिणामस्वरूप, NADP+ को NADPH + H+ में अपचयित कर दिया जाता है।
(These electrons are moved downhill to a molecule of NADP-. As a result, NADP+ is reduced to NADPH + H+.)

(This transfer of electrons, starting from PS II, uphill to the accepter, down the electron transport chain to PS I, excitation of electrons, transfer to another accepter, and finally downhill to NADP+ is called the Z scheme, due to its zigzag shape. This shape is formed when all the carriers are placed in a sequence on a redox potential scale.)
b. Splitting of Water (Photolysis):-
·        PS II से स्थानांतरित होने वाले इलेक्ट्रॉनों को प्रतिस्थापित किया जाना चाहिए। यह जल के विपाटन से प्राप्त इलेक्ट्रॉनों द्वारा किया जाता है।
(The electrons that are moved from PS II must be replaced. This is achieved by electrons getting by splitting of water.)
·         PS II में जल विपाटन जटिल थाइलाकोइड झिल्ली के अंदर की तरफ स्थित होता है।
(The water splitting complex in PS II is located on the inner side of the thylakoid membrane.)
·         जल H+, (O) और इलेक्ट्रॉनों में विपाटित होता है।
[Water is split into H+, (O) and electrons.]
·         प्रोटॉन (H+) का उपयोग NADP को NADPH में अपचयित करने के लिए किया जाता है।
[The protons (H+) are used to reduce NADP to NADPH.]
·         ऑक्सीजन को प्रकाश संश्लेषण के उपउत्पाद के रूप में मुक्त किया जाता है।
(Oxygen is liberated as a byproduct of photosynthesis.)
·         PS I से हटाए गए इलेक्टोनों का स्थान लेने वाले इलेक्ट्रॉनों को PS II द्वारा प्रदान किया जाता है।
(The electrons needed to replace those removed from PS I are provided by PS II.)

Hill reaction (हिल अभिक्रिया):- It is the light-dependent reaction of photosynthesis in which water is split by light, releasing oxygen and transferring electrons to an artificial electron acceptor (instead of NADP⁺).
[यह प्रकाश-संश्लेषण की प्रकाश पर निर्भर अभिक्रिया है, जिसमें प्रकाश द्वारा पानी के अणु टूटते हैं, जिससे ऑक्सीजन निकलती है और इलेक्ट्रॉन एक कृत्रिम इलेक्ट्रॉन ग्राही को स्थानांतरित किए जाते हैं (NADP⁺ के बजाय)।]
> The Hill reaction was discovered by Robin Hill in 1937. It demonstrated that isolated chloroplasts can produce oxygen in the presence of light, proving that oxygen released during photosynthesis comes from water (H₂O), not carbon dioxide.

> Proved that the light reaction and dark reaction of photosynthesis are separate processes.

> Established that water is the source of oxygen released during photosynthesis.

> Helped explain the electron transport chain in photosynthesis.

c. Photophosphorylation:-
·         कोशिकाओं द्वारा ATP के संश्लेषण को फॉस्फोरिलीकरण कहा जाता है। यह माइटोकॉन्ड्रिया और क्लोरोप्लास्ट में होता है।
[The synthesis of ATP by cells (in mitochondria & chloroplasts) is called phosphorylation.]
·         प्रकाश की उपस्थिति में क्लोरोप्लास्ट में ADP से ATP के संश्लेषण को फॉस्फोरिलीकरण कहते हैं
[Photophosphorylation is the synthesis of ATP from ADP in chloroplasts in presence of light.]
·         यह 2 तरीकों से होता है: अचक्रीय और चक्रीय।
[It occurs in 2 ways: Non- cyclic and Cyclic.]
i. Non-cyclic photo-phosphorylation:-
Ø  यह तब होता है जब दो प्रकाश तंत्र एक इलेक्ट्रॉन परिवहन श्रृंखला के माध्यम से एक श्रृंखला में कार्य करते हैं, (पहले PS II और फिर PS I) जैसा कि Z स्कीम में देखा जाता है।
[It occurs when the two photosystems work in a series, (first PS II and then PS I) through an electron transport chain as seen in the Z scheme.]
Ø  यहां, ATP और NADPH + H+ संश्लेषित होते हैं।
(Here, ATP & NADPH + H+ are synthesized.)
Ø  यह एक अचक्रीय प्रक्रिया है क्योंकि PS II द्वारा खोए गए इलेक्ट्रॉन्स वापिस इसके पास नहीं आते हैं, परन्तु NADP+ पर जाते हैं।
(It is a non-cyclic process because the electrons lost by PS II do not come back to it but pass on to NADP+.)
ii. Cyclic photo-phosphorylation:-
Ø  यह stroma lamellae में होता है जब केवल PS I कार्यात्मक होता है।
(It occurs in stroma lamellae when only PS I is functional.)
Ø  इलेक्ट्रॉन प्रकाश तंत्र के भीतर गति करता है और ATP संश्लेषण इलेक्ट्रॉनों के चक्रीय प्रवाह के कारण होता है
(The electron is circulated within the photosystem and the ATP synthesis occurs due to cyclic flow of electrons.)
Ø  ग्रेना के लैमीली में PS I और PS II है। स्ट्रोमा लैमिली झिल्लीयों में PS II और NADP रिडक्टेज का अभाव होता है।
(The lamellae of grana have PS I & PS II. The stroma lamellae membranes lack PS II and NADP reductase.)
Ø  इलेक्ट्रॉन NADP+ पर पास नहीं किया जाता है, परन्तु इलेक्ट्रॉन परिवहन श्रृंखला के माध्यम से पुन: PS I जटिल पर वापिस चला जाता है।
(The electron does not pass on to NADP+ but is cycled back to PS I complex through electron transport chain.)
Ø  यहां, केवल ATP को संश्लेषित किया जाता है (NADPH + H+ नहीं)
[Here, only ATP is synthesized (no NADPH + H+)]
Ø  चक्रीय फोटोफॉस्फोरिलीकरण भी होता है जब केवल 680 nm से ऊपर की तरंग दैर्ध्य का प्रकाश उपलब्ध होता है।
(Cyclic photophosphorylation also occurs when only light of wavelengths beyond 680 nm are available.)
d. Chemiosmotic Hypothesis:-
·         यह क्लोरोप्लास्ट में ATP संश्लेषण की क्रियाविधि की व्याख्या करता है।
(It explains mechanism of ATP synthesis in chloroplast.)
·         ATP संश्लेषण थायलेकोइड झिल्ली के पार एक प्रोटॉन प्रवणता के विकास से जुड़ा होता है।
(ATP synthesis is linked to development of a proton gradient across thylakoid membranes.)
·         जल के अणु का विपाटन झिल्ली के अंदर की तरफ होता है। इसलिए प्रोटोन थायलेकोइड्स की गुहा में जमा होते हैं।
(Splitting of water molecule takes place on the inner side of the membrane. So the protons accumulate in the lumen of thylakoids.)
·         जैसे जैसे इलेक्ट्रॉन्स प्रकाश तंत्रों के माध्यम से गति करते हैं, प्रोटॉन्स को झिल्ली के पार पहुँचाया जाता है।
(As electrons move through the photosystems, protons are transported across the membrane.)
·         ऐसा इसलिए होता है क्योंकि प्रोटॉन्स को निम्नलिखित कारणों से स्ट्रोमा से हटा दिया जाता है: -
(This happens because protons are removed from the stroma for the following reasons:-)
Ø  प्राथमिक इलेक्ट्रॉन ग्राही झिल्ली के बाहरी तरफ स्थित होता है और अपने इलेक्ट्रॉन को H-वाहक पर स्थानांतरित करता है। इसलिए यह अणु एक इलेक्ट्रॉन परिवहन करते समय स्ट्रोमा से एक प्रोटॉन को हटा देता है। जब यह अणु झिल्ली के भीतर की तरफ स्थित इलेक्ट्रॉन वाहक पर अपने इलेक्ट्रॉन को स्थानांतरित करता है, तो प्रोटॉन झिल्ली के अंदर की ओर (गुहा में) में छोड़ दिया जाता है।
[The primary electron accepter is located towards the outer side of the membrane and transfers its electron to an H carrier. So this molecule removes a proton from the stroma while transporting an electron. When this molecule passes on its electron to the electron carrier on the inner side of the membrane, proton is released into the inner side (lumen side) of the membrane.]
Ø  NADP रिडक्टेज एंजाइम झिल्ली के स्ट्रोमा की ओर स्थित होता है। PS I से आने वाले इलेक्ट्रॉनों के साथ, प्रोटॉन्स NADP+ को अपचयित करने के लिए आवश्यक होते हैं। इन प्रोटॉन्स को स्ट्रोमा से भी हटा दिया जाता है।
(The NADP reductase enzyme is located on the stroma side of the membrane. Along with electrons coming from PS I, protons are necessary to reduce NADP+. These protons are also removed from the stroma.)
·        इस प्रकार, स्ट्रोमा में प्रोटोन्स की संख्या में कमी हो जाती है, जबकि गुहा में प्रोटोन्स का संचय हो जाता है। यह थायलेकोइड झिल्ली के पार एक प्रोटोन प्रवणता बन जाती है और गुहा में pH में कमी आ जाती है।
(Hence, protons in the stroma decrease in number, while in the lumen there is accumulation of protons. This creates a proton gradient across the thylakoid membrane and decrease in pH in the lumen.)
·         प्रोटोन प्रवणता के टूटने से ऊर्जा निकलती है।
(Breakdown of proton gradient leads to release of energy.)
·         एंजाइम ATPase के F0 के ट्रांस-झिल्ली चैनल के माध्यम से झिल्ली के पार स्ट्रोमा में प्रोटोन की गति के कारण प्रवणता टूट जाती है।
(The gradient is broken down due to the movement of protons across the membrane to the stroma through the trans-membrane channel of the F0 of the enzyme ATPase.)
·         ATPase (ATP सिंथेज) में दो भाग होते हैं: -
[The ATPase (ATP synthase) consists of two parts:-]
Ø  F0:- यह झिल्ली में धंसा होता है और एक ट्रांस-झिल्ली चैनल बनाता है जो झिल्ली के पार प्रोटोन्स का प्रेरित विसरण करता है।
(It is embedded in the membrane and forms a trans-membrane channel that carries out facilitated diffusion of protons across the membrane.)
Ø  F1:- यह थाइलेकोइड झिल्ली की बाहरी सतह पर निकला रहता है। प्रवणता के टूटने से उत्पन्न ऊर्जा F1 कण में एक गठनात्मक परिवर्तन करती है। यह एंजाइम कोATP अणुओं को संश्लेषित करने में सक्षम बनाता है।
(It protrudes on the outer surface of the thylakoid membrane. The energy due to breakdown of gradient causes a conformational change in the F1 particle. It makes the enzyme to synthesize ATP molecules.)
Ø इस प्रकार, रसायनपरासरण में एक झिल्ली, एक प्रोटोन पम्प, एक प्रोटोन प्रवणता और ATPase की आवश्यकता होती है।
(Thus, chemiosmosis requires a membrane, a proton pump, a proton gradient and ATPase.)
Ø  ऊर्जा का उपयोग प्रोटोन्स को झिल्ली के पार पम्प करने के लिए किया जाता है, ताकि एक प्रवणता या थाइलेकोइड झिल्ली के भीतर प्रोटोन की उच्च सांद्रता बनाई जा सके।
(Energy is used to pump protons across a membrane, to create a gradient or a high concentration of proton, within the thylakoid lumen.)
Ø  ATPase में एक चैनल होता है जो वापिस झिल्ली के पार प्रोटोन के विसरण की अनुमति देता है। यह ATPase एंजाइम को सक्रिय करने के लिए ऊर्जा देता है जो ATP के निर्माण को उत्प्रेरित करता है।
(ATPase has a channel that allows diffusion of protons back across the membrane. This releases energy to activate ATPase enzyme that catalyzes formation of ATP.)
