Properties of trans genes (पराजीनों के गुण):-
1. Variability (परिवर्तनशीलता):- Transgene expression in plants is highly variable, even among plants independently transformed with the same construct, there is also no guarantee that primary transformants showing strong transgene expression will give rise to progeny with the same characteristics.
(पौधों में पराजीन अभिव्यक्ति अत्यधिक परिवर्तनशील होती है, यहां तक कि एक समान पराजीनों द्वारा स्वतंत्र रूप से रूपांतरित पौधों के बीच भी इस बात की कोई गारंटी नहीं है कि सशक्त पराजीन अभिव्यक्ति दिखाने वाले प्राथमिक रूपांतरक समान लक्षणों वाली संततियों को उत्पन्न करेंगे।)
2. Stability (स्थायित्व):- The production of transgenic plants with stable, high-level transgene expression is important for the success of crop improvement programs based on genetic engineering.
(आनुवंशिक अभियांत्रिकी पर आधारित फसल सुधार कार्यक्रमों की सफलता के लिए स्थायी, उच्च-स्तरीय पराजीन अभिव्यक्ति वाले पराजेनिक पौधों का उत्पादन महत्वपूर्ण है।)
3. Factors affecting transgene expression (पराजीन अभिव्यक्ति को प्रभावित करने वाले कारक):- Many factors may be responsible for variable transgene expression:
(परिवर्तनशील पराजीन अभिव्यक्ति के लिए कई कारक उत्तरदायी हो सकते हैं:)
i. Tendency for exogenous DNA to undergo rearrangement prior to integration
(समावेश से पहले पुनर्व्यवस्था से गुजरने के लिए बहिर्जात DNA की प्रवृत्ति)
ii. Position effects
(स्थिति प्रभाव)
iii. The effects of transgene copy number

Interaction and expression of trans genes (परजीनों की अंत:क्रिया व अभिव्यक्ति):-
1. Insect Resistance (कीट रोधिता):-
cry gene transfer (cry जीन स्थानांतरण):- The cry gene is found in the plasmid of Bacillus thuringiensis (Bt) bacteria. This cry gene makes Crystal protein which is insecticidal.
[Bacillus thuringiensis (Bt) जीवाणु के प्लाज़्मिड में cry जीन पाया जाता है। यह cry जीन Crystal प्रोटीन बनाता है जो कीटनाशक होती है।]
Ø The cry gene was isolated from the plasmid of Bt and integrated into the cotton genome to form a transgenic plant called Bt-cotton.
(cry जीन को Bt के प्लाज़्मिड से निकालकर कपास के जीनोम में समाकलित कर ट्रांसजैनिक पौधा बनाया गया जिसे Bt – कपास कहते हैं।)
Ø This Bt - cotton is resistant to boll worm.
(यह Bt – कपास boll worm के प्रति रोधी होती है।)
Ø Crystal protein perforates the larva's alimentary canal leading to its death.
(Crystal प्रोटीन लार्वा की आहारनाल को छिद्रित कर देती है जिससे उसकी मृत्यु हो जाती है।)
2. Virus Resistance (वायरस रोधिता):-
cp – gene transfer (cp – जीन स्थानांतरण):- The coat protein is transferred from the TMV virus to the tobacco plant.
(coat protein जीन को TMV वायरस से निकालकर तंबाकू के पौधे में स्थानांतरित करते हैं।)
Ø The RNA of TMV virus is not come out in the presence of cp-protein. Due to which the tobacco plant becomes resistant to mosaic disease.
(cp – प्रोटीन की उपस्थिती में TMV वायरस का RNA बाहर नहीं आ पाता है। जिससे तंबाकू का पौधा मोजेक रोग के प्रति रोधी हो जाता है।)
Ø Other examples (अन्य उदाहरण):- Tomato, Alfalfa, Beetroot, Potato (टमाटर, Alfalfa, चुकन्दर, आलू)
3. Seed Protein Quality (बीज प्रोटीन गुणवत्ता):-
SFA8 gene transfer (SFA8 जीन स्थानांतरण):- In sunflower seeds, the SFA8 gene produces a protein that contains an excess of sulfur-rich amino acids methionine and cysteine. While the proteins present in pea seeds lack methionine and cysteine amino acids.
(सूरजमुखी के बीजों में SFA8 जीन ऐसी प्रोटीन बनाता है जिसमें सल्फर युक्त अमीनो अम्ल मेथिओनिन व सिस्टीन की अधिकता होती है। जबकि मटर के बीजों में उपस्थित प्रोटीन में मेथिओनिन व सिस्टीन अमीनो अम्लों की कमी होती है।)
Ø Therefore the SFA8 gene is transferred to pea seeds.
(इसलिए SFA8 जीन को मटर के बीजों में स्थानांतरित किया जाता है।)
4. Gene Silencing (जीन साइलेंसिंग):-
Slow ripening tomatoes (धीरे पकने वाले टमाटर):-
Ø PG (Poly Galacturonase) enzyme digest the pectin. Due to which fruits become soft and fluffy quickly and ripen quickly.
[PG (Poly Galacturonase) एंजाइम पेक्टिन का पाचन करता है। जिससे फल जल्दी नर्म व पिलपिले होकर शीघ्र पक जाते हैं।]
Ø Anti-sense genes are integrated into the genome of tomato plants against genes that code PG enzymes. This inhibits the expression of PG gene and the tomato fruit ripens late.
(PG एंजाइम को कोड करने वाले जीन के प्रति Anti-sense जीन बनाकर टमाटर पादप के जीनोम में समाकलित कर देते हैं। जिससे PG जीन की अभिव्यक्ति रुक जाती है और टमाटर का फल देरी से पकता है।)
Ø Antisense gene made against PG gene inhibits its expression, which causes tomato to ripen late and can be easily exported.
(PG जीन के प्रति बना Antisense जीन इसकी अभिव्यक्ति को रोक देता है जिससे टमाटर देरी से पकता है व इसे आसानी से Export किया जा सकता है।)
5. Male Sterility (नर बंध्यता):-
Producing male sterility (नरबंध्यता उत्पन्न करना):-
Ø Flavonoid are essential for maturation of pollens. If we stop its formation then male sterility can be created in the plant.
(Flavonoid परागकणों के परिपक्वन के लिए अनिवार्य होते हैं। यदि हम इसके निर्माण को रोक दें तो पादप में नर बंध्यता उत्पन्न की जा सकती है।)
Ø The CHS (Chalcone Synthase) enzyme is required for flavonoid synthesis.
[CHS (Chalcone Synthase) एंजाइम Flavonoid संश्लेषण के लिए आवश्यक होता है।]
Ø Anti-sense genes are integrated into the plant's genome against genes that code CHS.
(CHS को कोड करने वाले जीन के प्रति Anti-sense जीन बनाकर पौधे के जीनोम में समाकलित कर देते हैं।)
Ø Absence of a flavonoid causes pollen to become inactive. The plant becomes male sterile.
(Flavonoid नहीं बन पाने से परागकण निष्क्रिय हो जाते हैं। पौधा नर बंध्य हो जाता है।)
6. Biochemical Production (जैव रसायनों का उत्पादन):-
• PHB (Poly Hydroxy Butyrate):- It is prepared from Acetyl CoA. From this bio-degradable plastic is made.
(इसका निर्माण Acetyl CoA से होता है। इससे जैव अपघटनीय प्लास्टिक बनाई जाती है।)
• The transgenic plant is developed by isolating the phb - B and phb - C genes from the Alcaligenes eutrophus bacterium and transfer to Arabidopsis thaliana. Now PHB is obtained from this plant.
(Alcaligenes eutrophus जीवाणु से phb – B व phb – C जीनों को निकालकर Arabidopsis thaliana में स्थानांतरित करके ट्रांसजैनिक पादप बनाया जाता है। इस पौधे से PHB को प्राप्त करते हैं।)
